STUDY OF ANTIOXIDANT ACTIVITY OF THE PHYTOREMEDY «TIGROVIY GLAZ – ORTHO” |

Reactive oxygen species (ROS) in high concentrations can damage or kill cells and tissues. Their action is at the bottom of great number of diseases (e.g., tumor and cancer, chronical inflammations) and lead to complications in pathology (e.g., diabetes millitus). Opposite, ROS in low doses can regulate intracellular reactions and promote cell survival. Nature compounds and their compositions are potential exogenic scavengers of ROS and other free radicals. To consider it necessary deep scientific analysis to use the phytoremedies in practical apply to improve health. Phytoremedy “Tigroviy glaz – ortho” was disigned at the Scientific Production Association “MEDICOMED” (Moscow, Russia) as panto-restorative, adaptogenic and antioxidant complex. Compounding of bioactive additions “Tigroviy glaz” was taken into account of biological activity of standardized dry plant extracts from Bergenia crassifolia, Arctium Lappa, Betula verrucose, bark of Salix alba and complex of amino acids (collagen hydrolysate). Task of the work was to study antioxidant activity of phytoremedy “Tigroviy glaz – ortho”. The preparation properties was estimated on the in vitro experimental models by chemiluminescence analysis using a chemiluminometre “CHEMILUM-12” designed at the ICB RAS (Russia). The phytoremedy “Tigroviy glaz – ortho” was examined unfractionated human blood of health donor-volunteers, isolated granulocytes and cell-free enzyme reaction. The preparation decreased level of the activated ROS production in the whole human blood in dose-dependent manner. Since the neutrophils are the most active to produce ROS in the whole blood, the remedy in different dilutions (from 103 to 106 times) was tested on the isolated cells. Pretreatment of the cells with the “Tigroviy glaz – ortho” significantly decreased spontaenous level of ROS production and depressed intensity of the respiratory burst induced by chemotactic peptide N-formyl-Met-Leu-Phe (fMLF) – product of the bacteria degradation. Effect of the preparation depends on the dose. The effect of the specimen on the NADPH oxidase activity of the cells ranged from low decrease (dilution 106) to complete depression (high concentration, dilution 103). Then the plant preparation was tested on cell-free enzyme reaction “xanthine – xanthine oxidase”. Superoxide-anion radical is product of this reaction. Its dismutation gives rise to other oxygen intermediates such as hydrogen peroxide, synglet oxygen, and hydroxyl radical etc. The plant preparation inhibited the reaction in dose-dependent manner similar to superoxide dismutase. Thus, the phytoremedy “Tigroviy glaz – ortho” demonstrates antioxidant activity in the whole blood, on isolated cells and in the enzyme reaction. We conclude that the product “Tigroviy glaz – ortho” possesses a very high antioxidant activity in the doses reached in dilution more than 100000 times. Antioxidant action of the preparation directs to superoxide-anion radical being initial form for other reactive oxygen intermediates. We suggest that the application of the phytoremedy “Tigroviy glaz – ortho” could be efficient in diseases complicated by phagocyte dysfunction and abundant unregulated ROS production.
