INVESTIGATION OF THE AXILLARY AND MESENTERY LYMPH NODES UNDER EXPERIMENTAL CANCEROGENESIS OF THE BREAST CANCER IN THE CONDITION OF CHEMOTHERAPY |

Structural and cellular investigation of the mammary gland, axillary and mesentery lymph nodes is carried out under conditions of chemotherapy of induced cancerogenesis of the mammary gland.
Experiment was conducted on 60 Wistar rats-females, 3 months of age, with initial weight 220-250 g. N-metil-N-nitrozourea (MNU) was used as a cancerogenic agent. A mammary gland tumor was induced (under ether narcosis) by the described technique (according to Dzhioev F.K., 1989) by means of hypodermic injections of MHU in the area 2 of the mammary glands in the basis of the right forward finiteness in a dose of 2,5 mg on a rat in 0,2 ml of water for injections, once a week within 5 weeks. 3 experimental groups were formed: 1. Intact animals; 2. Induced cancerogenesis of the mammary gland; 3. Induced cancerogenesis of the mammary gland under condition of chemotherapy. CMF scheme (Cyclophosphan/Methotrexate/5-Fluorouracil) was applied as chemotherapy treatment of the animals with breast cancer. Euthanasy of the animals was carried out under ether narcosis. Axillary and mesentery lymph nodes were taken for histologic research.
In regional (axillary) lymph nodes under condition of chemotherapy of experimental tumor the following changes were revealed in comparison with non-treatment group: reduction of quantity of metastatic tumoral cells in structural components of lymph nodes on more than 80 %, structural signs of activation of a cellular link of immunity (the area paracortical zone is increased), blast transformation processes of lymphoid line cells (the number of the small lymphocytes in germinative centres is increased) and proliferation of plasmatic line cells (the number of plasmoblasts in brain substance is increased), decrease of macrophage reaction and reduction of the area of sinus system.
In mesentery lymph nodes under condition of chemotherapy, unlike group without chemotherapy, metastatic tumoral cells are revealed only in brain substance where their quantity is considerably reduced. Structural signs of activation of a cellular link of immunity (the area of thymus-dependant zone is increased) are marked, activation of blast transformation processes of lymphoid line cells and proliferation processes of plasmatic line cells is revealed, reduction of quantity of macrophages and increase – of neutrophils is marked.
Thus, the received experimental results testify that breast tumor recourse under condition of specific chemotherapy leads to decrease in quantity of metastatic atypical cells in the sine of lymph nodes. Structural-cellular transformations in regional (axillary) and peripheral (mesentery) lymph nodes under condition of chemotherapy of breast tumor testify to activation of their barrier-detoxication function, to stimulation of cellular and humoral immunity links.
