PROSTATE AND REGIONAL LYMPH NODES UNDER CONDITION OF EXPERIMENTAL TUMORAL GROWTH AND TREATMENT BY CISPLATIN AND NEW ANTINEOPLASTIC AGENTS ON THE BASIS OF PALLADIUM SALTS |

Detailed studying of the transformations in the regional lymph nodes in experimental tumor growth in the prostate can help to understand a role and functions of the lymph nodes in the pathological process. The analysis of structural changes in the lymph nodes in experimental tumor growth under condition of treatment by platinum and palladium preparations can allow to find differentiated approach to a choice of an antineoplastic preparation.
The work purpose is to reveal structural transformations in experimental prostate tumor (18 days of tumor growth) and pelvic lymph nodes under conditions of parenteral treatment by preparations of «Cisplatin» and new antineoplastic agents on the basis of palladium salts, developed on chair of the general chemistry of Novosibirsk State University, Institute of inorganic chemistry of the Siberian Branch of the Russian Academy of Science.
Materials and methods. The experimental model of malignant tumor growth in the prostate was created by inoculation of dissolved cellular stamm of ascite Ehrlich’s tumor in the prostate parenchyma in СVА males-mice. Experimental groups: intact animals; 18 days of tumor growth; 18 days of tumor growth and application of pharmacological preparation «Cisplatin»; 18 days of tumor growth and application of the substance on the basis of palladium salts. Medical products («Cisplatin» and substance on the basis of palladium salts) were entered daily (in a dose – 2 mg/kg of weight of an animal, intraperitoneally) once a day within 5 days, since 6th day after inducing of tumor process in the prostate.
Results of research have shown that under conditions of experimental tumor growth in the prostate and of introduction of new substance on the basis of palladium salts, in comparison with Cisplatin (pharmacological preparation), expressed transformation of tumor tissue is revealed (expressed necrosis of tumor node, considerable growth of connecting tissue, with preservation of muscular fragments, increase of number of less differentiated atypical cells), marked increase of stroma-parenhyma – on 62,5% and nucleo-cytoplasmatic indexes– on 3,5% is found.
In pelvic lymph nodes under condition of antineoplastic therapy by substances on the base of palladium salts, in comparison with Cisplatin, structural signs of humoral immunity activation were revealed (B - dependent zone is increased by 41,5%, brain – by 59%, lymphoid, plasmatic immature cells numbers and mitotic cells are increased), oppressions of the cellular immunity links are found (the paracortical zone is reduced). Activation of lymphopoetic and decrease in transport function of regional (pelvic) lymph nodes testify to reduction of antigene loading on the lymphatic region, to safety of barrier-detoxication functions of the lymph nodes.
Thus, the obtained data allows to make the conclusion that under conditions of experimental tumor growth in the mice prostate and introduction of new substance on the basis of palladium salts more expressed destruction of tumor tissue is observed (necrosis of tumor node on peripheries, increase of connective tissue component and number of high-differentiated cells in the tumor stroma), in comparison with group with monocorrection by Cisplatin, on the background of structural reorganisation of the regional lymph nodes, with signs of structural activation of transport function and plasmatization of lymphoid parenchyma. Results of research allow recommending substance on the basis of palladium salts for creation of new antineoplastic preparations.
